Close tubes and incubate on ice for minutes.
Heat Shock:
- Place tubes into water bath for 50 seconds at 42°C
- Back on ice for 2 minutes
Remove from ice.
Add 0.25mL of LB-Broth into each tube, using a transfer pipette.
Incubate at room temperature for 10 minutes.
Transfer 0.1mL from the tubes onto the appropriate, labeled plates.
Use sterile loop to spread evenly across agar.
Stack plates upside down and place in incubator at 37°C.
