Easy How-To Steps:

Label 2 eppendorf tubes:

+pGLO and -pGLO

Put 0.25mL of transformation solution (CaCl₂) into each, using a transfer pipette.

Place tubes on ice.

Use innoculation loop to pick up a bacteria colony

from starter plate. Immerse loop into +pGLO tube.

Place back on ice.

Sterilize loop by placing into a flame.

Repeat step 3 with -pGLO tube.

Put 10μL of plasmid DNA using a micropipette

ONLY INTO +pGLO
