Disease is a Personal Matter
DESCRIPTION
“Medicine is a science of uncertainty and an art of probability,” said Sir William Osler (1849–1919), a father of modern
medicine. After one hundred years, his statement still remains true, because regimens and diagnostics are developed
according to population-averaged statistics in spite of large human diversities. As a result, on average only one out
of four patients responses to the cancer chemotherapy. Artificial intelligence (AI) can identify an input-output relation
with a mechanism-free approach. We trained neural networks with experimental data and discovered that the effica-
cy is related to the drug doses through a parabolic response surface (PRS), which is governed by a quadratic poly-
nomial. The AI-PRS equation holds true for the cell line, animal and clinical tests. With a small number of calibration
tests on the patient, we can determine the personalized coefficients of the AI-PRS equation and customize the opti-
mal drug-dose for that specific patient. Applying this AI-medicine platform to treat an 85 old man with metastasized
prostate cancer, the PRS platform can guide us to home in the sweet spot of customized drug-dose combination
with high efficacy and low toxicity.
We have clinically demonstrated successful personalized therapies for cancers, infectious diseases, and organ trans-
plants. With this indication agnostic AI-PRS platform, it can determine the optimal drug-dose combination and dy-
namically adjust the doses for about 30 different disease models
