medium . Dilutions were applied in a volume of 100 μL in each well of the microdilution plate . Thuja occidentalis extract was added in quantities of 25 μL , 50 μL , 75 μL , and 100 μL . The plates were covered and left for 24 hours at 37 ° C . After 24 hours , optical density was measured at 540 nm using an ELISA reader . Tests were performed in triplicate for all samples . Suspensions of strains in Brain Heart Infusion medium were used as positive controls . The microdilution method in the culture medium is one of the simplest methods for testing antimicrobial sensitivity . The technique involves testing double dilutions of the analysed antimicrobial agent in a liquid growth medium distributed in 96 wells plate .
Conclusions
Based on our research , we can deduce that the extracts , particularly when used at the initial concentration tested , exhibit an inhibitory impact on Gram -positive bacteria . The inhibition of Gram-positive bacteria escalates in the following sequence : Cl . perfringens < S . pyogenes < S . aureus < B . cereus < L . monocytogenes . The observed outcome of Thuja occidentalis golden smaragd suggests its potential as a future candidate for natural products possessing antimicrobial properties . Our discoveries might pave the way for utilising Thuja occidentalis golden smaragd blends in diverse applications , including developing dermatological treatments for bacterial infections and the food industry . These sectors face significant challenges due to the growing resistance to conventional commercial antimicrobial products .
