Enzymaster : One-stop solution for next generation biocatalysis

Since 2013 Enzymaster provides a one-stop solution for the development and commercialisation of innovative and sustainable enzyme catalysis technologies . Our portfolio includes enzyme identification , enzyme panel screening , smart enzyme engineering , process development , enzyme preparation by fermentation , and biocatalytic manufacturing from kg to tonne scale . In addition to these services , we also offer general enzyme kits representing various enzyme classes , as well as customised enzyme kits that fit to the individual biotransformation needs of our customers .

Natural enzymes are often not readily applicable for industrial processes for reasons like lacking robustness under operating conditions , insufficient activity , and product and substrate inhibition . The fast discovery of novel enzymes using bioinformatic methods and directed evolution facilitated by computer aided enzyme engineering have enabled overcoming these hurdles .
Key to the successful identification and development of commercial enzymes are the proprietary BioEngine ® and BioNavigator ® platforms . BioEngine ® is our branded directed evolution toolbox with integrated computational enzyme engineering . Based on the computational results , we construct smart and small libraries of enzyme variants , which can be conveniently screened via HPLC or GC , or MS to characterise a comprehensive range of desired properties for these variants under process conditions . This enables us to reach the biocatalytic process targets in typically only three to six rounds of combined in silico and experimental directed evolution which corresponds to a timeline of four to eight months .
The BioNavigator ® is a set of state‐of‐the‐art methods for computational enzyme identification , in silico activity and selectivity prediction and stability calculation under process conditions . We offer bioinformatics driven hot spot identification , focused combinatorial library design , and in silico loop design . AI and structure‐based
recombination algorithms are applied for in silico pre-screening to enhance the combinatorial sequence diversity coverage beyond traditional random and site-saturation mutagenesis approaches , thereby increasing the sequence space coverage and identifying all important sequence areas . This allows us to shift most of the screening efforts from the laboratory into the computer .
We have already developed several industrial processes employing a diverse array of enzyme classes in our R & D pipeline . Newly developed processes include the production of API intermediates for Ubrogepant and Pregabalin , drugs that are used for the treatment of migraine , and for epilepsy and neuropathic pain , respectively . The production of the Ubrogepant intermediate was achieved via dynamic kinetic transamination using an engineered transaminase whereas the production process of the Pregabalin intermediate is based on an engineered hydantoinase .
While pharmaceutical intermediates still remain one of the main targets for biocatalytic processes utilising engineered enzymes , we have also developed processes aiming at the production of polymer building blocks or food supplements from renewable resources . Examples are the production of the nylon precursor cadaverine from bio-based L-lysine via an engineered decarboxylase and the production of the keto nutrition ingredient ( R ) -1,3-butanediol from bio-based rac-1,3-BDO via engineered keto reductases . ●
