There’s a big difference between the movie and comic-book versions of Deadpool: However, both came to a conclusion that he was in fact born a mutant. Yet in what sense?

X-Men, such as Wolverine and Colossus (Whom are considered as mutants) possess mutant genes ever since birth; however, they wern't mutants from the start, they only possessed the genes that can ptentially cause them becoming mutants - a new huma specie other than the homo-sapiens . Mutants like Spider-Man and Daredevil get their powers after an event triggers genetic mutation – most famously, Spider-Man was bitten by a radioactive arachnid.

In Deadpool, Wade was injected with a syrum that did not activate any of his ordinary genes like with captain america, but it was manfuactured for the sole purpose of "turning on" his mutant gene. That means he was born with the potential to develop regeneration powers, in other words, he was born with the mutant genes that would give him his regenerative powers, waiting to be activated.
